Coastal zones are important ecosystems with high productivity and biodiversity. Monthly surveys were conducted from April 2018 to March 2019 (winter season data were absent) to examine fish diversity in Hakodate Bay, Hokkaido, Japan, using environmental DNA. Water samples from two different rivers (Kunebetsu River and Moheji River) were collected at the surface along transects 0, 125, 500, 1000, and 2000 m from both river mouths. Samples were also collected from the middle and bottom layers at 1000 and 2000 m from both river mouths. Both river mouths seemed to have less diversity than the coastal areas. Species richness of the Kunebetsu River was higher than that of the Moheji River only at the river mouth. In the coastal area, the Moheji River showed the highest species richness. However, species richness between the rivers showed significant differences, neither by month nor by distance. This result indicates that forest coverage and human activity did not substantially affect fish species richness in Hakodate Bay. When the fish community was divided into three groups, namely river mouth, surface water, and middle/bottom water, the species composition between the two rivers appeared to be quite different at the river mouths and became more similar offshore. |
